In the de-ess/breath control video, it's mentioned that those 2 and de-plosive would commonly be combined into a module chain for workflow purposes. Is the idea that these could be all applied to the entire piece of audio at the same time? I've been warned against applying de-plosive to anything but short individual instances of it happening.

Yes, you could create a Module Chain of common vocal processors and apply them all at once. You could still choose to apply that Chain to smaller sections of a longer audiofile, one section at a time, if you're concerned that the settings you have for De-plosive might be overly aggressive in certain parts of the recording. Either way you should probably test the settings first and still check the recording after the processing is applied just to be sure there are no unacceptable artifacts from the processing.
At some point if you find yourself using that Chain on similar recordings regularly with no issues, then you could probably apply the processing with more confidence, but it's always a good idea to play through the audio afterwards just to be sure.
